I'm running a Predator one-shot coming up, but I'm also curious for Assassins as well:
If a party knows that something dangerous is in the area but doesn't know what it is, they would almost certainly set a watch for a long rest. Is it possible to surprise that PC on watch? They know there is danger, but don't know where or what it is.
A person can't maintain high vigilance for hours, ready for all possible occurrences. Roll perception against stealth, and grant surprise as you see fit.

Or, give a distraction (sees cloaked figure creeping into houses) when the PC on watch is least likely to wake the others up.
