Karl, I forgot that if you get bit, you also take poison damage. Of course, this is silly for a dwarf, but let's roll it out just for the fun of it. Roll a DC 11 CON save or take 9/2=4 poison damage. If you save, you take 2 poison damage.
Vondal, the drow is visible because he tried to cast a spell on Karl. Your arrow hits, and the drow turns his angry glare on you.
I would say if you move to one of the angled square, you can grab the handle as well, so Nif could then release it on his next turn. But as you will be holding it at an angle, your grip will not be as good as standing right in front of the door (I'd essentially lower the DC a little).

Nif ponders the meaning of "I'll kill the dwarf!", before he releases the door handle mutters a few words under his breath, waves his hand and a hand appears and holds onto the door handle, continuing to hold it shut. He then steps back to O9, and crouches using the stairs as some protection, keeping his crossbow at the ready and says. "Why should I believe that you can kill the dwarf from there, without sight of him, perhaps you need to get out to kill him."

I think I need to, the spell has verbal and somatic components. If I can cast the spell and just transfer the holding to the mage hand, I will do that.

Ah, I forgot about the somatic component. Yes, unless you put away (or drop) your crossbow, you'd have to release the handle to perform the spell. I'll go with your original posting as you had all of the information (some of which I was forgetting).

The dwarf looks in to see a short, dark-bearded human male in robes holding the room-side of the handle in one hand and a glass staff in the other. The room is lit by a lantern, and a dwarf is sitting in a chair and tied up in the corner behind him. The human looks surprised that he the door opened.
Nif, you still have your action and bonus. What would you like to do?

Nif drops his crossbow and lunges at the man, attempting to grapple him and pin him to the ground. Unfortunately, the human is a little more nimble than the rogue expects, and Nif is unable to grab hold.

Nundro stands and moves away from the desk.

The (red) and (green) spiders attempt to bite Karl (Attack: 21 Damage: 4; Attack: 20 Damage: 11), but neither is able to pierce his barbarian's skin. The (pink) spider sprays webbing at Vondal. (8) Unprepared to defend himself from an attack from the ceiling, the monk finds he is restrained by the webs.
Vondal, you're up, and you're restrained. You can break out with a DC 12 STR check.

By the time Vondal starts to relay Nif's words, even if somewhat in futility, the words turn into half spoken curse as the webs descend upon him. "No.. nonono... nooo! Seeing Nundro make a break for it after all this time sparks finally sparks something in Vondal, completing the circuit and eliciting a scowl. "Kinda trapped at the moment, Brother Nif!" He called back while struggling to line up a shot. And when that proved too daunting a task, he settled moving his hand just enough to point at Nundro. "By Marthammor... I hope I'm right. Ignisium!" From the dwarf's finger tips shot a small ball of fire aimed right for Nundro.
He then calls out, "I know what you are, Faker!"He called out after the spell misses its mark, hoping to at least force Nundro to confirm his suspicion at last."Uncle! Hammerer! Hurry those spiders. Nif's in trouble. And I... can't... free myself yet." As if to emphasize his point, Vondal made a token effort to try and pull free but to little avail.

@Whatwall: Understood. He was surprised he was finally able to open the door, but not that surprised. He knew someone was on the other side and holding the door. If you had said that you released the door and then immediately jumped inside to grapple whatever was on the other side (chancing that it was something you'd want to grapple), I'd probably have gone with the surprise and given advantage. But since you were actually releasing the door with the intent of conjuring mage hand to hold it closed, you were pretty much just as surprised the door opened as he was. Therefore, no advantage. I appreciate you bringing up the question just in case. It never hurts to ask.
His attention turned from the fallen drow to the fifth dwarf in the room, Karl moves out of melee with the two giant spiders. Both attempt to bite him. The first lunges, but Karl casually swings his shield, and it bangs loudly into the creature's face. The other, however, manages to give him a small bite.

Finally, what lies on the floor is another hairless, genderless and apparent naked gray-skinned humanoid like what you saw in the castle.
In the separate room, the bearded human attempts to charm Nif.
