I would personally enjoy seeing everything stripped down to the mechanics. Toss out any lore and anything based on lore (spellcasting ability and such) so people can put in their own lore. I would also like more optional features and other customization options.

A choose-your-own-adventure tutorial, like in Red Box. Introducing D&D to new players is a serious challenge, and a story tutorial that introduces the mechanics one by one would be perfect. I honestly have no clue why this isn't already right on the first pages of the PHB.

If we are just talking 5E there really isn't a whole lot more to ask for. The game is fully matured at this point with tons of content. However for 6E they really need to rework ability scores. That should be the main focus. It is dumb how people consider certain ability scores as useless "dump" stats. That shouldn't exist, every ability should be equally viable to invest in for any character, like if I want an intelligent barbarian who fights with brain instead of brawn that should be perfectly viable, or dexterity wizard who is a quick fingered spell slinger.
The lackluster "optional" feat system we have could be abolished and the feats spread out as feat trees tied to various ability scores to support this. When you get an ability score increase you get your bump AND a feat from one of these trees if you meet the pre reqs.
How you are distributing your feats and bumps would be a major driving force behind your character build, as much as your class, and every ability would be viable for anyone.
Various species are evil, because their gods made them that way, as per the traditional lore.
Players' backstories do not play such a key role. Whatever happened to the 3rd son of loving parents from a small village simply striking out on his own for fame and fortune?
An overhaul of the Feats. Either balance them (Lucky is banned at my table, Observant can ruin any suspense in a game), or remove them entirely. I think Healer is a great Feat, but no one ever takes it, because so few tables play by actual RAW when it comes to players being at 0 HP. So certain Feats should simply be removed or altered.
A streamlining and balancing of the sub-classes. 18 subclasses of Cleric, but only 8 for Sorcerer? 10 and 10 would be reasonable.
Much more onerous limitations on multi-classing. If a player MC's, bring back hard caps on levels they can get in one class.
A DMG that's actually a guide for dungeon masters. A PHB that's a better handbook for players. A MM that's a proper manual on monsters.
I love crunch, whether to use as is, cannibalize for homebrew or throw out and replace with houserules, but all the books (the DMG most of all) could do a better job of covering how to play rather than what there is to play. For the MM we have Volo's and Mordenkainen's at least and it's not that the PHB doesn't try, but there's room for improvement.

Included in that would be the impact of magic items and the relative power of a 6 player group versus a 4 player group would be hugely helpful. It is not linear.
Also include in the PHB AND DMG many of the standard questions with answers about the RAW of Invisibility, Hiding, Perception, Investigation etc. Crawford has cottage industry of tweeting what the rules mean. If examples of RAW with the most ambiguous cases were provided, that would go a long way towards clarity. If a DM chooses to ignore RAW, so be it. But at least new DM's know what is the baseline.
Also, a section in the DMG about customizing monsters so not only are metagamers hung out to dry, but so experienced players can be surprised and challenged.

Well, DDB has a search feature, which is sometimes useful, depending on whether you get the search term right and whether or not the word appears in multiple books in different contexts. It's not as good as having an index to a single book, because the search is, as far as I know, not able to be restricted to just one source, and you might know the source, just not the actual page.
It would also help if WOTC had put things in logical places, but I can't tell you how many times I've not been able to find something using the chapter headings because things are not in the chapter where I'd expect them to be. For example in the DMG, the discussion of creature size and its meaning comes under the heading of "Using Miniatures," as if the only time size would matter in a game is if you are using miniatures. "Using miniatures" is not the first place I would think to look when wanting to find the game definition of "large size..."

I think a lot of these ideas are good for newcomers to the game... a lot of suggestions seem to be focused on making the published books better at teaching players and DM's the things that aren't extremely obvious. And I agree that it would be good to have more content focused on that... but honestly, I feel like I know the system pretty well so while that would all probably be pretty useful to most people, it's not really what I would want, personally.
I think I'd like to see another new class added to the game. I can't help but think about how exciting it was when Artificer was added to the game, and all these cool new character concepts and even changes to whole settings that came with them. Unfortunately, I don't really have a good idea for any new classes that couldn't reasonably be just a subclass to an existing class. I suppose I'd like something maybe like a "Shaman", which essentially just be a Druid but without the baggage of Wildshape. I don't dislike Wildshape, necessarily, but it feels kind of weird that if you want to be a dedicated caster with a very nature-focused spell list you need to also be a shapeshifter. It's kind of a weirdly specific skill attached to the class.
I would like a Psion class, a book dedicated to magic that provides more spells and comprehensive magic item creation rules, and a book dedicated to Martial classes that gives new weapon and armor choices as well as abilities to add to their options in combat.
Just whatever you would like to see.

Having had my beginnings with 4e, I'd like to see a fully fleshed Warlord class come out which buffs/plays off their allies.
And a Nentir Vale book, now that I think about it. Pretty sure that'll never happen, but it'd be nice.
