I'm wondering if Tree Stride can be used for vertical movement? Can I enter at the base of one tree and exit onto a branch 30 feet up in that tree or another? I can't see how a climb speed would apply, by the description of how the ability is used. Can I stride into a tree and exit into a cave that connects to it's root system?
you wouldn't be able to pass through the roots into a cave. But that's because the roots (at least under any normal circumstance) would have the size limitations required to tree stride.
Otherwise Tree Stride does not dictate how you enter and leave trees other than the need for it to be able to physically fit your body at the entrance and exit points, that it's the same kind of tree as the one that you enter through, and that it's within 500 feet of where you enter said tree.

The thing is, at least in my campaign setting, there are big magic trees everywhere. SKT has one with roots wider than normal trees.

Well, you could make a dungeon hidden under the roots of one of those trees that has been causing sinkholes in town. That might be fun.

I did make room for your custom setting situation by saying under normal circumstances. your custom setting would not be a typically normal circumstance.
Though Giant tree's is something i have used in some of my own custom settings as well. But for the both of us when we do it. That kind of becomes a DM Fiat type thing that makes it possible that is the exception that proves the Rule when it normally doesn't work because we've created a situation where it does purely at our tables.
So as long as I observe the distance rules, movement rules, tree size rule and exit from a tree other than the one I entered, it shouldn't matter what part of the tree I exit from (assuming the the DM also agrees to this ruling)? I love the idea of a hidden cave under an ancient tree that only a skilled ranger can enter! I'm also imagining ways that this could be broken in a place like the Feywild or Happy Hunting Grounds or any other place with giant trees.

I’d allow it as a DM.

Tree stride goes from 1 tree to another. Not to another part of the same tree
