No, Silvery Barbs gives the target disadvantage on the one roll, and that's it. If you have Extra Attack and Silvery Barbs is used to make you re-roll your hit on the first attack, you can still make your second attack, which is as normal, but you now know that character who Silvery Barbed your first attack can't do that to your second one because that character already used their reaction.
Also, the Paladin's Extra Attack feature is tied to the Attack Action, not only melee attacks. I think you're mixing that up with the Paladin's Divine Smite, which it can only add to melee attacks.
