So Dark Sun is clearly the one being announced next as it's been pretty overtly hinted that it will be. The question is what the second one will be? It could be Spelljammer or Planescape. Despite what the article says, my money is on Planescape.

Mearls talking Psionics and the Half Giant character at SoME does hint really heavily at Dark Sun. I would LOVE a Spelljammer book but I would expect that Eberron maybe higher on the list or maybe Dragonlance, since they brought back the Minotaur again for testing.
I think there should be a book (Perhaps called D&D Multiverse) that has some info for each of Greyhawk, Eberron, Spelljammer and Dark Sun. It would at least be something.
I think the two settings that can be played as early as this year will appear first in some kind of UA or playtest PDFs available on dmsguild or some such, and they'll gauge interest that way. As such, they could be just about anything. Put me down for Dark Sun and Eberron. As for the book they are announcing in July, I think it will be a Planescape/Sigil sourcebook/adventure with a revamp of Spelljammer to make it fit more smoothly with the cosmology.
I'd love to see Nentir Vale/Points of Light get some love, but I'll understand if it doesn't. I'd rather them leave Greyhawk alone. I already ignore just about everything after the 1983 boxed set. I'd be all right with some Krynn love.

Stewart was quoted as saying that people could start playing as early as this year, so I am not so sure of that. In fact, it's possible they have produced something to be released through dmsguild because they don't plan on doing traditional print books for them any time soon. This could save money and please fans of those settings at the same time.
Assuming what comicbook.com reported is accurate, I’m agreeing with what A_Stray_cat said, and I’m guessing it will be something like this:
The releases will be a setting summary/conversion guide that will realease on dmsguild.com - no physical books.
And coinciding with each settings release they then allow self published content created for that setting to be sold in the dmsguild (right now that is only forgotten realms and I guess Barovia)
Just a guess, but I’m not getting my hopes up. I think they’ll let the dmsguild creators put out a bunch of content, and then they can see what has traction.
WOTC seems to have been very careful about who can tinker with their settings, so they might keep this all in-house. But I don't think so. Maybe it's my tinfoil-hat-wearing self, but farming out work to Guild Adepts, dmsguild's experiments with hardcover printing, and this announcement of new material beyond WOTC's usual 5e output seem related to me. We know that WOTC has had to do more with less for quite a while, even though they have made a couple significant hires in recent times. They have made some meaningful partnerships to expand the brand and be able to steward what other people produce. Being able to leverage community members' natural creativity has been a boon for them.
If I'm right, and these start off as print-on-demand products through dmsguild, I'm trying to convince myself that it might not be too bad. A small sandbox with details along the lines of Curse of Strahd, a small amount of text aligning the setting to the chargen books and a broad paintbrush of the history, and license to knock yourself out creating whatever - those things might be better than a full-on setting book that I have to whittle at to make the world I want to run in. Additive vs. subtractive setting creation. That's one of the things I both found useful and frustrating about Nentir Vale. There is so little background that what I didn't like I could easily ignore, but what was there was the beginning of a skeleton I could hook meat to. But finding it spread throughout 4e materials (I discovered Nentir Vale late because I skipped 3rd-4th editions, going straight from 2e to 5e.)
I would be very disappoint in a dmsguild release instead of an actual book.
Unfortunately, I'd prepare for disappointment. From everything they have said (including Nathan Stewart's comments from the beginning of the year about their "big plans" for DMs Guild, I'm absolutely convinced it'll be a DMs Guild release maybe like the Plane Shift products. If it's not, then it's gotta be into next year.
Now, with Print-on-demand as A_stray_cat notes, a physical book might be possible. But, no, I really, really doubt it's going to be a physical book at Amazon or your FLGS. Everything they have said points to DMs Guild, and all of the statements have been very careful to avoid indicating it might be another hardcover through normal distribution.
(Also, we should probably keep in mind that ALL of this speculation is based on 1 single reporter. Although WotC hasn't denied anything or tried to clarify any misquotes/misunderstandings.)
They have to make the right selection if they want my $. Most of the oddball settings will not open my wallet.

I'm expecting a book with a bunch of setting conversion material like the Eberron UA in it, but for several different settings. And I expect it'll be about the size of SCAG.
With all the talk about Psionics something for Dark Sun sounds like a good chance, I don't really see a need for a Half-Giant race when all you need is the goliath. i think the biggest issue is the Attribute range, When DarkSun appeared stats were 3 to 18 but in Dark Sun they went over that, so maybe allow stats to push to 24 or so. They might even have to expand generation methods for the setting. I can see it being an actual book for the fact of Adventures League.

FYI: Stats were 5-20 (rolled 5d4), which does have a higher average (12.5 vs 10.5) and higher max & min, but it also is more likely to be closer to the average than 3d6. So mostly it sounded very impressive but in actual play was only somewhat more powerful, especially since every group I played in was already 4d6 drop lowest anyway.

Dark Sun and Seplljammer are my preferred, but this would be fantastic! Also, I'd love for them to release updated material on the rest of Toril. I used to love al-Qadim.
