Will I need to buy them a second time to use them here? And what if I'm using a VTT like Roll20 or Foundry? Do I need to purchase again? Lots of questions need to be answered!

You're going to need to buy them again for a different site to use them there. This is unchanged from how it worked before.
If using Roll20, there's another app called Beyond20, which connects DDB to Roll20 (and Foundry and Discord). It's possible to buy the books once here on DDB and play via Roll20 by using Beyond20.

The question is a technical one. Will the API be provided to those developers or is there going to be a VTT lockout?
There's no official API to begin with. While it's not impossible we'll see one post-release of WotC's VTT, I expect it'll remain status quo, where the stuff that gets built to use DDB is gonna break from time to time because things get changed.
(The only reason we might get a stable API after the VTT is that Maps and the VTT are likely both using the same interfaces, at which point they need a more controlled change process to keep them in sync.)
