In theory yes. However, when it was first launched, there were complaints that the skills weren't relabelled appropriately and you had to do a lot of stuff manually, negating the point of using DDB.
Whether that's still the case I don't know, I haven't heard anything in a while...which could be because they've fixed it, or it could be that very few people have bought it recently.

Former, yes, with some caveats depending on how the features are supported. You can make a character for the LOTR RPG using the D&D Beyond character builder.
Latter, no. The character builder only presents information using the DDB character sheet interface, or the default 2014 character sheet PDF if you export it.
