The navigation checks must always be made, whether they are made by the guide or a character. I let one helping the other in the check if both have proficiency in Survival.
If the guide KNOWS where they are going, they have been there before, then there is no real need for a navigation check to be made. That is the purpose of a guide, after-all.
If the guide is "familiar with the area" but is not familiar with the specific location, then yes, they must use a navigation check.
WOTC recently released a UA (called into the wild)that deals specifically with this question. You should look into it.

If the party has a guide do they make navigation checks, does the guide or is there just no navigation check made?
I'm also curious how DM's went about introducing the party to Omnu.

For Omu, I went with Saba N'baja in Orolunga.
I let the party choose who makes the check and let they use the help action if more than one person has survival.
My players took three guides so that they could cross check the information the guide give them...their so paranoid and I don't understand why.
