Is there any mechanic that allows/prevents Daunting Roar from expanding its AOE if you use it while enlarged.
In my head you go from a 1x1 5ft space to occupying a 2x2 10ft space and the radius of daunting roar is 5 ft; so wouldn’t that make its radius 4x4 20ft radius?

It will expand, as all AOEs do that have the emitting creature as their point of origin. Such AOEs emit from the creature's edges, not its center.

As quindraco explains, effects originating from a creature are measured from the edge of the space the create occupies.
The Leonin Daunting Roar affects creatures of your choice within 10 ft. In gridded combat(where everything is squares/cubes) when a Leonin uses Daunting Roar, it affects a 25ft by 25ft square with the space the Leonin occupies being the 5ft square in the very center. If the Leonin was under an effect that made them Large size then Daunting Roar would affect a 30ft by 30ft square with the Leonin occupying the 10ft by 10ft square in the center.
