First things first, you can't make an item that allows for a single casting of a specific spell. Items get up up with charges, and then spells get added with a set amount of charges they use. In other words, your item, if set up like most magic items that give spells, would need 5 charges, with Hunter's Mark using 1 charge as a first level spell, and Locate Creature using 4 charges as a 4th level spell. BUT... this means that (strictly per the item created) it would look like the player could just cast Hunter's Mark 5 times, using 1 charge each time.
Alternatively, you could give the item 2 charges. Locate Creature requires 2 charges, and Hunter's Mark requires 1. This means that the PC can either cast Locate Creature 1/day, or Hunter's Mark 2/day.
To do this:
Under "Additional Information", check the box for "Has Charges", qty: 2, reset: long rest
Under "Spells", min/max charges on HM 1, min/max on LC 2
You can either give the item 2 charges and make each spell use 1 charge and put in the description that it is once each or make 2 different items with 1 charge and 1 spell.
Being able to set separate limits is one of the missing features DDB needs to add to support certain official content as well.
