our longtime DM has a master tier subscription and has been sharing out content for our games. If I want to step in as the DM is that possible? Do i just start a new campaign and have the master tier friend just enable sharing? Or are there additional steps i need to be aware of?

DDB has no idea who is actually DM'ing. Also, if you use the same DDB 'campaign' the limit is on number of players, not on number of characters. So if all the same people, there should be room for their characters in there too

Some corrections:
D&D Beyond will place the campaign creator as DM of the campaign so yes, the OP would need to make their own campaign to be DM. As DM they can edit private and public notes and have extra options if using Avrae bot in Discord.
There is a limit on characters. Player limit is 12, IIRC and character limit is 99.

Depends on what you mean by 'DM.' If you mean 'view and edit characters in the campaign OOC' then yes, who owns the account matters. If you mean 'Run the campaign using Avrae or Roll20' then no, since neither of those care who owns the account. Or if they do, not seen any sign of them so far.
