I'm currently working on a campaign where one of the BBEGs is a half-elf turned vampire. I have a character sheet set up for him as a regular half-elf, but I want to use the "players as vampires" method from the Monster Manual to determine his vampire stats. Is there any way I can do this?
You mean do so on D&D Beyond? No, there currently isn't a way to create a character and lair or vampire, lycanthrope (what I think used to be called "template" in prior editions) over it.
Moreover if you were using the encounter builder on DDB, you can't drop in character sheets as monsters "on the other side" of the encounter anyway, so if that was your leaning, you'd have to build a stat block anyway.

You should be able to either homebrew a vampire monster to match it, or homebrew a feat that adds the vampire actions/resistances to a PC sheet. If you run into issues, post in the Homebrew forum to ask for help. A quick glance shows that there are multiple homebrew feats already created by others and published for vampire traits. (I didn't look at each one, so can't recommend the best one for you)
I had one of my players transform into a vampire in a previous campaign, I didn't follow the monster manual guidance, instead, using Grim hollow vampire transformation that brings more option and the ability to make you vampire transformation evolve.
It is something you can homebrew with feats, and it works well, I've also used that for the lich transformation for a BBEg and it worked just fine!
Yes, I did misspeak in that there are homebrew work-arounds. My response was speaking of whether there was a way to "afflict" a PC with vampirism within the official system (homebrew lacking quality control and more prone to error than stuff build by the devs). Homebrew isn't too hard to implement, adopt or even design on your own ... but it does require some practice if you've never done it before (though the homebrew section of this forum provides great help).
There is the question as to whether it's necessary at all to go through a whole character sheet for a vampire NPC. My recommendation would reflect a prior content where you homebrew the NPC has a monster derived from the MM Vampire stat block. Some people do like to work out "major" NPCs via character generation. I personally find that labor excessive an inefficient where you can basically drop in whatever powers you want into the monster and not worry about conforming to character race and class stats which are already being bent in the first place to accomplish what you want.
