In the lightly forested area the trees seemed... unwilling? to grow near the river. Is that the same in the heavily forested area? i.e. is there a buffer of spongy riverbank between the trees and the river?
Yes, there is. There's like a 15ft gap between the water and the forest. However in the deep forest section, it has algae and moss growing in that section (normal sized. Not the giant algae and moss).
Endurance will (because I had totally not read the bit about the gap before) fill a bowl of river water (from his mess kit) and pour it on some giant moss/tree. Also, did the amazing pup come from the direction of the river or not?
Endurance does that the giant moss/tree doesn't give a shit. It probably enjoyed the water. So yeah, the river water is not an amazing "Anti-Scary-fungus" fluid. There must have been some other reason for trees not growing there and it's not the water itself. Also the dog didn't come from the river's direction. Now that you traveled upstream it seems the puppy had come from the direction of the dense forest... where the trees are infected even worse.
Alright so everyone seems to want to head east so that would take you to where you see faint lights. Please note that these lights are too far away from any city on the map so it must be something else.
Regardless you head east and soon come to the grassland. All the grass has been replaced by the moss but now you know where you are. You are definitely at the place where the road was supposed to fork. The map shows a hill to the north and you can see it in the dim light. Now as to the source of the light, scattered about in this place are strange purple mushrooms that glow. That's where the light is coming from. These glowing mushrooms are peppered in the whole grassland. Though you can't make out details of the hill, the glowing mushrooms show the silhouette of it even though it is still a bit far away. However, in spite of the hill's presence marking you at the right place, there is no road here.
The pup barks as per usual. You've been traveling for quite a while. What do you do?
(Aside from the pup, during our travels have we seen any indication of fauna? Whether that's insects to slap at, birds in the trees/moss, game to hunt?)

You begin heading east but as you do so the puppy begins growling. You then notice it begin splitting into two from the middle. The cub's body opens up and green slimy tendrils emerge. You notice the inside is covered in a black moss and algae. Looks like the cub was infected but it was on the inside. Spreading 4 long algae-like tendrils, the small body stands up over your heads. The two shorter tendrils remain aimed at you like the claws of a mantis. Hard teeth-like bones line the split in the body from where the tendrils emerged. The beast doesn't make a howling or barking sound. Instead it makes a wet gargling sound as slippery plant fluid drips down the split like drool.
Roll initiative.
Also take 9 Sanity damage or half on a DC15 WIS save.

You did not encounter insects yet but there were a two or three birds that you saw. These birds were also infected by algae, fungi and moss.
