If I update my hero tier to master tier do I lose my character creations?
You will not lose any characters you've created by upgrading your subscription tier.
I should also ask on the flip side. If I decide master tier isn't for me and I decide to go back to hero would I lose anything? I paid Hero anually. IS master tier pro rated?

Both hero tier and master tier subscriptions provide unlimited character slots. So nothing will change in that regard by switching tiers.
If you cancel your subscription, you'll be asked to select 6 characters to remain active. The others will remain on your account in an inactive state.
