For Feats, in the Prerequisite section, the only Proficiency options you can select from are armor types. Please add other proficiency options (e.g. weapon types).
Something that would be pretty cool is giving items the chance to give special actions to the player. Like, for example, i made an item that gives the player a reaction to take damage instead of a companion. However, i had to manually add the action to the player, because the item can't have it.
Ability to modify range on weapons, and add the Ammunition property to weapons that don't have it already.
For example, i want to make a sniper crossbow that has a range of 300/1200 feet. I can't do it in any way. This is quite frustrating to be honest with you.
I would like to be able to have limited use feats and race traits that don't reset on Short/Long rest, but with working boxes. As for now it says you have 4 uses/special, and you can't bar those uses. Also, have the option of "Set X score to Y" work for races. For example, if a player is affected by Lycanthropy or Vampirism i have to create a custom item, instead of putting a feat/modified subrace.

One example would be adding a Proficiency or LevelValues as prerequisites: Proficiency (pop-up list) at least 3 (Fixed Value), or Level (pop-up list) at least 5, and so on...
What do I mean with Template? A list of features (Feats I previously have homebrewed separately), that can be added to characters without using a Class, a Race, a Subclass, or a Subrace.
Templates usually come or emerge by training or events that either improve or alter your normal Class, Subclass, Race and Subrace abilities, without needing to substitute them.
A Chosen of Mystra (or any named deity of your choice), for example, gains special ability and traits available only to them, impossible to achieve by any of the indicated above character heritage nor path.
I know actually we can add any number of features one by one, but due to the incredible amount of homebrew feat the community has created, I feel everyone would say "Thank You" if there would be a way to avoid reinvent hot water every time. Simply, add up in a template the list of homebrew features you want you character gain, and assign that template to it.
A brief note about me: I spent 15 years coding this kind of things for PCGen, since the very early release of it. I see that the developing team of DDB is running, but the speed and destination are far away from the expectations of people using that tool for so many years. You definitely need to ask us for directions, and follow our indications as soon as possible, or DDB will be surpassed by faster growing and more community-aware Character Management Systems, like MPMB Character Sheet.
I'm not sure if this is what you mean by "modifiers," but I'd like to see the d3 added as an option anywhere we have a dice roll pop-up menu during homebrew item and spell creation.
I'd like to be able to adjust a background spell list to make guilds akin to those in the Guuldmaster's Guide to Ravnika.
Also, the Actions and Spells sections on Races and Subclasses should be on everything. Races and Subclasses have gotten pretty awesome while Feats and Magic items are pretty awkward by comparison.

customizable spellcasting features such as:
-additional spell slots via feat or items
-adding a feat to a non-spellcaster that allows it to have spellcasting available (spell slots, spell slot level scaling, etc...)
I'll second this one. Especially the second part. The bonuses on magic items should really only effect that specific item.

Yeah, especially fighting styles. Or at least add the ones from UA like tunnel fighter and such...

Can the Spell Points rule variation from DMG be added as an option for home brew spell casters?
+1
This option can later be used when (if) the Mystic class is ever added.

Allow Homebrew Feats to provide Class Features of another class, like Fighting Styles, etc.
Homebrew class features that modify creature stats (like custom animal companion or wild shape).
Homebrew base classes.

I would like the ability to use the template of a homebrew variant, or subrace.
Item activation.
So how items which needs action or bonus actin to activate.
An activation checkbox when customizing things like AC or weapons/armor.
For AC this would make applying things like Mage Armor or Shield much easier to do.
For weapons, it makes appying things Blessing of the Forge easier.

We seriously need maths and formulas...

We definitely need Templates.

Edit: and d2 also
Recently returned to D&D after 20+ years.
Unapologetic.
I would like to have a homebrew modifier to adjust the maximum number of prepared spells.
A replace ability feature, such as Replacing Rage with another ability, not sure if that is possible
