So could a Kensei spend all of his Ki points for big boy damage in a single attack with either one of the abilities/the combination of the two?
You can combine these abilities, but they are limited. The former is a maximum of 1 ki point of 1d8 and the latter is up to 3 ki points for a maximum of +3.
If you can you are just doing it for the Hunter's Mark, you may consider going 2 levels of Warlock to get Hex along with 2 spell slots on a short rest. The extras for warlock aren't bad either. Getting a SCAGtrip can help with the damage during the lead up phase, too, at least while lower level.
However, Ranger will be easier to Multiclass to and the ability to get Archery fighting style would be beneficial for bow attacks.

Not to mention, with the Variant Class Features coming out, a 2 level dip into Ranger has other perks for a monk as well with both Canny and Roving having added perks in addition to Favored Foe and Spellcasting to boot.

If it's a ranged attack from a kensei weapon you can also stack Kensei's Shot for an additional 1d4 on top of all that.

The benefits of Sharpen the Blade last for 1 minute, you don't have to activate all these in the same turn.
