The party is a group of spies infiltrating a secret organization to that's trying to overthrow the king.
Tell them beforehand that one of the PCs will be randomly selected as a secret double-agent working for the secret organization. Their characters even know there's a mole.
Now the party is constantly investigating each other and trying to figure out how to deal with the mole.
Plot Twist: You lied. There was never a mole.
Probably would work fairly badly. But it's amusing to think of.
