Some rule benders will reference magic users and the use of two handed weapons in combination with spells like Shield to imply that the scenario is plausible for anyone. However they “forget” that the spell in question is designed to be a reaction, and hence does not cost the same amount of time as drawing a weapon, you could let go of the weapon with one hand and make the required gesture before the weapon position is unrecoverable. Anyway, a wizard casting shield will not be able to take an opportunity attack, as he has already used his reaction for the shield, hence the position of his/her weapon would not be an issue.
How do I set up a two handed attack with say a quarterstaff?
Thanks
If you are talking about in the character builder, it is automatic. It lists the one handed damage right above the two handed damage.

But how do I make it the default setting on the D&D Beyond site?
