If from our side, Duncan will go up to it, check the lock, knock on the door, and listen through it. PERCEPTION 10. The element of surprise is gone at this point anyway.

To the group: "Hey guys... There's something strange on the other side of this... We should tie that guy up first though." and Duncan will get his rope out and tie the mad scientist to a chair. "What's behind there?!" pulling the rope tight INTIMIDATION13 (can I use STR for INTIMIDATION? If not, take that score -3)
Elber makes sure to gag the alchemist as well, one can never be to careful with talky city folks, before taking position in a corner with a view of the chained door, arrow nocked and ready to loose. "As ready as ever."
Remembering something Landrae turns to Kai, "Kai, do you think you could garner any more information through charming him, before we proceed? Or shall we just forge on?

Jack asks his comrades to hold. “Let’s see if there is anything in this room that can help us destroy whatever is on the other side of that door. He did say ‘children’ so there could be two or twenty of them. I also suggest taking him” pointing to the old man, “into the hallway so they don’t free him.”
”Kai, help me search this room if you would for anything useful”
Investigation (w adavantage from Kai’s help): 23 Jack starts tossing the room. He’s looking for anything that can help damage these things, key for the lock, information, as well as hidden compartments. He will also search the old man.
Lastly he will move the candles away from the pentagram thing on the floor and scuff up the markings.
Landrae sees Jack move to the circle and candles, "Does anyone have much knowledge in the way of religion? Might those symbols or circle have had any significance?"
Jack says “I am ready to pick the lock once everyone is ready.”
JJ:Is there a way to leave the door chained after unlocking so it opens only partially? My idea is too limit what can come out of the door all at once.
"I was working in the lab, late one night When my eyes beheld an eerie sight For my monster from his slab, began to rise And suddenly to my surprise, He did the mash, he did the monster mash"
Landrae will give Elber her last bit of Bardic Inspiration.
"Is it worth trying to appear as if I was him?" Landrae says, pointing at who she presumes to be Fosa. "It might buy us a momentary advantage in the game. Speaking of which, are we playing hide and go slit throats, or should we just be up front about playing pin the arrow in the ooze?"
I will be off line for a bit so JJ feel free to have Jack open the lock when all is ready. I will leave someone to break the chain, stand on the other side of the room with my bow at the ready.

Jack is greeted by a soft click as the lock gives way to his expert touch. The gibbering within the room ceases, and all is silence for a few seconds. Then all hell breaks loose.
The chain shudders under the impact of some unknown mass and then snaps, and half a dozen gibbering horrors pour into the room. Pus and blood oozes from the numerous scabs and wounds they bear, and tongues hang grotesquely from open mouths. The alchemists voice, muffled but still audible, rises above the cacophony of chaos and horror in mad laughter.

The door is chained shut from your side. Approaching the door, Duncan can just make out a faint gibbering which chills him to the bone.

"I have no time for such foolishness... What say you friends, shall we see what Hosvi was so scared of?"
@Kevtr0n, Intimidation will be a skill on your character sheet, with your modifier on there but the modifier is off CHA though not STR.

After tying up the scientist, Duncan looks at Landrae, back at the door, and says "I'm ready. Let's see what's in there."

“Lady Luck is the only god I need.” Jack says not looking up at Landrae. He grasps something around his neck for a moment and continues his search.
Landrae, you come to the same conclusion as Duncan regarding the alchemist's current mental state.
Duncan and Elber, within moments you have the alchemist bound and gagged. He offers no resistance.
Jack, combing the room for useful items you find only a few empty glass bottles and dusty books.
The iron door on the far wall is both chained and locked.

Im assuming Jack is close to the door as he didn’t make to the opposite side of the room before the chain broke?
