I am trying to stack the Sentinel Feature with my Ancestral Guardian path on my Barbarian. Since my paths' level 6 feature allows me to use the reaction to reduce damage taken to another creature and the Sentinel Feature allows me to use my reaction to make a melee weapon attack against the attacking creature when it makes an attack to a target other than me. My Question is would be that since I will use my reaction to reduce damage once the target hits could I use the Sentinel reaction to hit back when the attack made fails? Almost giving me a reaction whenever an enemy attacks anyone else.
Also with Sentinel it can be more advantageous to use your reaction on your normal Opportunity Attacks to prevent an enemy from getting by you to the squishy backline in the first place.
I know you only get one reaction per turn, I guess i asked it wrong. If I got Sentinel I would have 2 possible reactions to make, so I would be able to choose which one i want to make that turn if it fits the reactions parameters. Can the Sentinel reaction be used when the enemy attacks and misses?

Yes. You can have several possible reactions (granted from one or more game features) but once you use one then you are out of reactions (until start of your next turn).
If someone attacks someone within 5 ft of you and you have Sentinel but have not used up your reaction, you can use your reaction to make a melee attack on them.
If you are raging and someone takes damage within 30 ft of you and you have Spirit Shield (from Barbarian subclass) but have not used up your reaction, you can use your reaction to reduce that damage by 2d6.
You can never use both the Sentinel and the Spirit Shield in the same turn.
Your reaction resets on your round, so if your initiative is 10, on init 11 you could use one of these abilities, in init 10 take your turn, and then use another (or the same) ability.
Spirit Shield is much more likely to grant a reaction, because of the range of 30 and the fact that it does not have to be an attack. It could be a save, or it could be an auto-damage feature such as Magic Missile.
Sentinel is one of the best feats around because it turns you into a wall that stops people from fleeing. No one really gives a crap about the attack, people take this to stop the Evil Bad Guy from turning around, dashing behind a curtain and escaping in a comically shaped villain themed vehicle. That crap gets really annoying really fast.
Yes, these two features combine pretty well... The Spirit Shield is only triggered by a successful hit on one of your allies, but Sentinel has multiple triggers that activate it... if an ally next to you is targeted by a creature within range of your attacks... if the attack misses, you can sentinel them in the face. If the attack hits you can use your Spirit Shield to reduce damage or, if the damage isn't too bad or if you know the ally can handle taking the damage, you still have the option to use your Sentinel counterattack.

Sentinel is triggered by an attack, not a hit or a miss, so it's probably resolved before the attacker even rolls to hit (I say probably because 5e isn't super clear about the order of action resolution).

You only get one reaction, and once you use it, you don't get it back until the start of your next turn, so no.
