Im about to play a Lizardfolk rune knight (i wasnt sure about what choice to pick and ended up rolling random class and race). Ill use the standard array, probably str 15 dex 10 con 12(14) int 14 wis 13 (14) and cha 8.
Im not sure about the feats/asi choices. I was thinking brawny at 4th for a +1 in str and expertise in athletics to make a good grappler, but after that im not sure abot taking the asi at 6 and 8 to boost str to 20 or maybe shield master to shove as bonus action.
A second choice would be to make him dex bassed and benefit from his natural armor, maybe two handed fight to benefit from the extra d6 to damage, but the str adv from the enlarge would be lost.

I'm currently playing a Rune Knight, and some of your ASI/Feat choices will depend on which runes you plan to use. If you're going to use runes that force targets to make saving throws(Fire & Stone), then you'll likely want to pump up your Int. Obviously pumping your attack stat is always a priority. There's nothing that prevents you from making a Dex-based Rune Knight. There are only two specific things that are Strength-focused: Invoking the Frost rune to boost your Strength by 2 for a minute, and the Advantage on Strength checks & saves when in Giant Might form. I usually try and get my attack stat to at least 18 as quickly as possible, I don't mind holding off on 20 for a while.
With just about every Fighter or Barbarian I play, Resilient(Wisdom) is one of my go-to feats at some point. You'll have Con saves covered, but being nerfed or even controlled as a result of a failed Wis save SUCKS OUT LOUD :)
Hmm the dex one would get better ace, and you are right about the wis saves, also he may be able to climb in the walls of the dungeon as a lizzard does with some ath checks. Maybe the build could be:
Str 14 dex 15 con 12 (15) wis 13 (14) int 10 cha 8
Archery fighting style
Brawny (dex, athletitcs) lvl 4, ASI dex +2 at 6, sharpshooter at 8, resillent (wis) at 12.
Athletics with adv, expertise, +2 str (8) AND storm rune disadv should be enough to grapple when needed as monsters dont have thay much atl.
I'm building one now for level 8. Using half orc 20/12/14/16/10/8 . Here's where it gets kinda spicy. I took the piercer feat @4th and added to strength to get to 18. The weapon I'm going with is pike for the piercing dmg ( Lance was also an option but ymmv). Took hill rune(resistance), cloud rune(those nat 20s deflection), and I'm debating between the frost rune (str buff) or the storm rune( gain adv by giving up reaction basically). This means if I crit I will be hitting for 4d10 + w/e is active every crit! This coming from the base 2d10 on a crit, then the extra d10 from piercer, then the other d10 from being a half orc. It nice.
I love the Brawny feat and I believe it’s a great addition to a dedicated grappler build alongside Giant Might.
Take a look at the new Unarmed Fighting style presented at the UA Class Features Variant, that will probably figure in the new Tasha’s book soon. It’s a great pick for grapplers.
I wouldn’t bother for Shield Master because you will have plenty of uses to your bonus action with the Rune Knight abilities.
