Any of the wamphyri from the Necroscope series, they put any of the vampires above to shame, and, given they are a parasitic creature that turns the host into a vampire, they can leave the host and move onto the next easily.

On note: Cain from WoD (assuming the lore to be correct) is up there with the Sacred Ancestor when it comes to vamps I DON'T want my players fighting. 😂. Nice suggestion though IamSposta for the Rosario + Vampire suggestion, despite it being an anime I give you credit for being someone cultured enough to remember the series, you have my upvote for that one.
As for a lot of "Live action" vamp characters... I kind of feel they don't quite measure up to the hype of other fictional vamps. Kudos to the Count Orlok suggestion though, definitely gonna keep that one in mind.
You could take inspiration from the World of Darkness though, have Cain in slumber, and the adventurers are trying to stop his resurrection. Could even have them in the middle of a vampire civil war, with one sect trying to revive there long lost sire believing he will help them conquer the planes, while the others understand they truth, the oldest of them put him into his endless sleep to begin with because he was going to destroy the planes to attempt to ascend as a true vampire god.

I'm going to have to disagree with you on Lestat not being the most powerful. After he absorbed some of the power of the Queen of the Damned, he became utterly ridiculous.

I fully support Cain, especially for what I see as a brilliant incorporation of classic real world mythology into a role playing game. The idea of "the first murderer" being the absolute original Vampire was just so awesome to me.

The most powerful vampire in the Dresden Files is probably Vlad Drakul, AKA Dracula's father, who casually manhandles Harry and a squad of his most powerful allies including Listens-to-Wind and River Shoulders. According to the author he's an actual god that was stuck in human form.
