1) Have something that prevents easy access to the object the party is looking for if they are trying to reach it by means of magic. This could perhaps be an antimagic field that is bound to the vault or to the entire casino. If you do this, I would think it would be a good idea to perhaps have the field be originating from a magic item or device inside the casino so that the party has the option to "turn off" the magical security as a possible side mission.
2) In addition to my first recommendation, I would have the vault be lead-lined so that the party cannot see into it with divination magic.
3) Obviously there should be plenty of security that works the casino floor as well as maybe mechanical traps along the main path to the vault (with possible secondary paths the party can uncover like air ducts and whatnot).
4) Since this is D&D the the owner has a "hoard" of rare items, perhaps the owner can also have a small pet dragon that sits in the vault. Due to the lead lining of the vault that I recommended, the party wouldn't know it is in there, so including other clues to hint at its existence would be good.
These are just some ideas I had come up with pretty quickly, and I hope they help. I had never really thought of doing a casino heist in D&D and it sounds like a very fun encounter/mini-dungeon

find some maps for casino layouts to use as battle maps of show the general layout for the recon I hope they do, a search on google images has loads for that.
I like this one from the Ultra Lux from fallout new vegas:
quite a few questions I would ask myself when designing it are:
How many ways in and out of the casino are there?
How many guards do they have and do they have the observant feat to catch thieves/cheaters?
Does the casino use chips like Las Vegas or do they only deal with betting in cold hard currency with Platinum Pieces for the high roller tables?
Is there sewer access?
Is it a front for the mob/thieves guild to launder money?
Is the vault where the rare items are located a fake one? Does the owner have them safely sequester else where?
Is the casino warded against divination such as sry, clairvoyance etc and does this just block any attempt or does it give a warning to the security along the lines of the Alarm spell?
Do the guards have access to Detect Magic? do they look over customers as they enter to see if they have any magical auras and then stop and search the ones that do?
Do they have tensers floating disks acting as elevators to get between levels or just stairs?
Does the Bank side of the Casino change money from copper to silver to gold etc and where is this held? In a vault? in a Leomunds Secret Chest?
What is the casino's reputation like/ are they known for being honourable and dealing fairly or are they thought to be cheaters or do people who win big mysteriously disappear along with their winnings?
Does the casino have an infernal patron? Where you can literally wager your soul for a wish spell?
Escape wise can they teleport out or does the item vault have an anti magic force field or even a beholder as the final guard or is the vault actually a dragons hoard with a dragon?
Is there stables and wagons or carriages the party can use to make a getaway?
Security wise if passwords or keys are required which members of staff have access to them?
If it all goes wrong and a blood bath ensues is there any local militia or law enforcement that would come to the casinos aid?
If time is of the essence for escape consider using an egg timer or hour glass and holding a series of skill challenges.

Another cool twist could be that the casino's security measures are so advanced, only the most daring and clever players even think about trying a heist. Speaking of challenges, I came across https://casinosanalyzer.com/casino-bonuses/richardcasino.com that offers Richard casino free chip, and it made me think of how tempting it is for players to try their luck when the odds seem in their favor. Maybe the owner of the casino has a similar setup, enticing players with promises of riches while secretly stacking the deck against them.
Love the idea of adding a magic-based challenge to the heist! Antimagic field is a solid way to block the usual tricks. Maybe have it tied to a high-stakes poker game where the magic item is the prize? The players would have to outsmart the house and get their hands on it first.
