So just want to make sure I'm not missing something, they can stack correct. making the character from medium to large and the large to huge, and extra d6 on one attack and then an extra d4 on every attack?
They do stack but best done in one direction. Your "Giant's might" will only make you large (If you are smaller then Large, you become Large). If you start large then you won't get bigger. Enlarge will boost whatever size it starts with.
If somehow someone gets to play a fighter up to 18th level it can even go up to Gargantuan. Though RAW creates a funny scenario!
"You learn how to amplify your rune-powered transformation. As a result, the extra damage you deal with the Giant’s Might feature increases to 1d10. Moreover, when you use that feature, your size can increase to Huge, and while you are that size, your reach increases by 5 feet."
So if Huge from the feature you get the 5-foot reach boost, but if you get enlarged to Gargantuan you lose that boost.
That's dope I hadn't even thought about that! Though the long-limbed bonus only applies on your turn. Mainly matters for opportunity attacks, including those from Polearm Master.
So you get to make reactions from 15 feet instead of 20, I'd say that's pretty negligible. In exchange you get the whack people from a range that is equal to most of the thrown weapons short range.
Another fun grey area with this; if someone plays as a character with the Powerful Build feature, can a rune knight technically grapple gargantuan creatures? That is a fun image.

"Powerful Build: You count as one size larger when determining your carrying capacity and the weight you can push, drag, or lift." and "The target of your grapple must be no more than one size larger than you and must be within your reach." They have no overlap.
All of that said, everything is up to DM discretion, even official Sage Advice rulings.

And +5 if you chose the lunging attack combat maneuver from the superior technology fighting style so can sometimes have a bugbear with a 25 ft. Reach
how tall would the character be is there a calculator to figure out when you increase size how tall you become
The rules don’t say. At my table we’ve been treating giants might like the spell enlarge and saying it doubles the height.
