The Sphere of Analihilation - has the damage of 4d10 per movement through a creatures square, so if it has 100 feet of movement it could move through the same square many times to the max of its movement as well as many others squares making this a very hard battle to win. But just confirming that is can happen it chew through a whole party rather quickly.
The sphere of annihilation can only move in a straight line and will only have a max movement speed of 50 feet (depending on controller's INT) without a talisman of the sphere which will make its max movement 110 feet (still depending on INT).
Moreover when it does pass through a creature's space, they make a DC13 dex save. They make better traps and area denial than weapons.

with enlarge upon it The Sphere of Analihilation should have a bigger, DC 15 save being 4 feet wide.

That is entirely house rules. Enlarge/reduce increases the size, but says nothing about DC in RAW.
