"Talien, I'll take him," Rees says, holding out her arms for the kobold.
Talien it order the offers hand and sets Meepo piggy back on Rees's shoulder. He picks Grom up and places him on his shoulders. Can you see Grom? (couldn't help it short stuff good thing you have blood shot eyes or the short pun wouldn't work because Talien wouldn't have said anything.)
Which direction does the party want to go? 49 E is unexplored as is 48 N.
Once we get out of the room, we'll set the poisoned ones down. (Mechanically, do they get another save, is this time-based, or what?).
Let's give these guys a minute to see if they can shake it off.
It looks like there are doors between all these rooms on the map. Is that accurate? If so,
Rees, go listen at the eastern door to see if you can hear anything moving. Don't open it, just come back and let us know what you hear. (door to 49 east, and we're in the lower 48 east)
Talien, see if you hear anything at the northern door. Same deal, just listen and report back. (door to 49 north). Then we'll decide which one to check out next.

As you leave the room the air is visibly clear of spores and seems breathable again...
DM Note: You can make an intelligence check if you want to see if you can deduce more info
Rees you hear an audible thrumming from behind the door. There is a visible glow emanating from under the eastern door. It reminds you of the glow you saw coming out of the holes earlier in the day...
Straight INT? Can I counter offer with a Medicine check, assuming we're talking about the party's health and reactions to a poison? LOL
Your initial question sounded like you wanted to know more about the area of effect. You’re welcome to make a medicine check if you’re trying to determine their health statuses.
Shake coughs up the spores and groans in pain. "I feel like an ogre hit me." He wobbles on his feet a bit before looking to where they are planted on the ground. "I'm afraid this could affect my effectiveness for the foreseeable future friends."
Talien carries Grom on his back over to the door and returns to Alten and reports "there is no noise or sign of activity beyond that door" He continues to hold Grom on his back as he reports back.

Nice roll.
You look everyone over and can see the fresh(er)[still in a dungeon lol] air is already starting to help the coloration of everyone’s skin. The party seems to be recovering quickly.. the health of those who were poisoned should return to full by the hours end (duration of poisoned condition is 1hr). You do not believe there will be any lasting effects from the spores, but deduce that prolonged exposure might have a more devastating effect — possibly even death. It might be best to avoid the vines should you come across more.
You guys should be ok in a bit. Do we push on or wait until you feel better? I do think if we encounter more of those vines, we need to hold our breath and move on if at all possible.Maybe burn them, but I'm worried about smoke on top of poison spores.
OOC: push on or wait for them to recover? It won't count as a short rest since we just took one. We'd just be killing time and hoping we don't end up in a random encounter. I'm willing to go either way.

"There's something behind this door,"Rees reports back to the group. "A sort of thrumming sound and a glow like the holes we say earlier. Maybe better not to go there until everyone's feeling a bit better?"
I was thinking on a longer term basis, while we moved on. The map doesn't indicate another way out, so if we could block or jam it somehow, we wouldn't have to worry about what's in there coming up behind us.
But it's not a bad idea while they recover. Talien, would you please?

I think this was only a footnote waaay back at the beginning of the game when you first entered the citadel.. but most of the rooms have interconnected airshafts, so that everyone doesn’t suffocate.
Edit:
Ventilation. All keyed areas contain an adequate air supply. The air is renewed from countless cracks leading to the upper open cavern and the surface. These cracks are too small for any but creatures such as giant rats to navigate.

What was your door made out of Rees? Talien will ask before he moves to stand against the door.
To that matter what was Talien's door made from?
(Also Grom if you want down you might say so, Talien will still be holding you in place in his shoulders)

They're both your typical oaken door with an iron ringer handle found throughout the citadel.
