So, for example, a L2 character with a Wisdom of 16 rolls a Wisdom (Perception) check. Normally, they would just roll the d20 (we'll say they get 15) and add their Wisdom ability modifier (in this case, since they have a Wisdom of 16, that's a +4) for a total of 19. However, if they're proficient in Perception, they can then add their Proficiency Bonus to it. At L2, that's +2, so their actual score is 21.
Expertise is exactly the same, except you double your Proficiency Bonus. In this case, it would mean that they got a 23 instead.

Great explanation, but one glaring error I cannot ignore. If you have a 16 Wisdom, that is only a +3 bonus.

and as other's have stated, that wonderful (P)roficiency is great. Getting to add that number really shows off what your character knows. and for expertise, you can double it! Rogues and Bards get expertise a few times in their careers, which makes them great skill junkies.

I created a character at 1st level; it's a rogue. I have a couple of questions that I can not seem to answer on my own.
I know that skills marked with the letter "P" means I'm proficient in them.... What does the letter "E" mean?
I think it means expert or expertise, even if I'm right, can someone give me an example of how each is used during game play?

Since the post above, while good, didn't answer your question directly. P is proficient. E is you have Expertise.
