I suggest before you go too far in planning that you devote some time to watching Colville's Running the Game series. It wont' give you any advice around the 7 sins but it gives lots of good general advice.

To add to it:
As long as everyone is having fun, even if you’re not strictly following the rules, you’re doing it right.
If you cannot find a rule in about a minute, make a ruling, let everyone know that’s what you’re doing, and tell them you’re gonna look it up before next session.
It’s okay to make mistakes, everyone makes mistakes. That’s the best way to learn how to avoid them in future.
More specifically, reread chapters 7, 9, and 10 of the PHB. The vast majority of the stuff you will use almost every session is in those three chapters.
Thanks for the updates man and it seems you got a lot more done and honestly I am in the love with the idea that sloth is the strongest but is so lazy to do anything about it. You can for the greed make them give them up magic items or if their backstories are involved make them give up a valuable trinket. There is a lot to do with greed there but you are definitely have some good ideas cooking.
I would make the actual adventuring/goals for the players to get hold of 7 Virtues, manifested as objects or beings, to counter the Sins and make the BBEG battles possible or easier. Working with the resistance groups would be key to that.
Since I messed up and chose "reply" instead of "quote" (twice...) (this is my first time using forums here.. :$), I'm going to make these useful.
If I choose Wizard and Monks for Sloth and Envy, then I might have to make custom creatures for them. Does anyone have any advice on making custom bosses? Balancing, Levels, etc.? Any opinions/advice would be greatly appreciated.
One suggestion is to create the character through the 'Character Builder' using the 'Randomize' option and then from there edit the character to your liking but it at least can start you off with a template for each of your Class-based characters. I do this a lot for my games when I need a more developed enemy and I could change the level as needed to scale the character to what would be appropriate for the players.
