Dain restrains his urge to rush in, waiting for Margret to check the path for them. He has Sabastian out of sight, as he doesn't want so many familiars flitting about just now.
The halfling will take out her poison and smear her bolts with the sticky substance [DM - Belladonna Poison]. Then she'll carefully move the dead orc and drag it over the trapdoor to slow down anything coming up through...
She will get The Prick ready for unpleasantness. When Margret gives the signal, she will pop up and shoot the enemy present in the NE section below.

After a second of dizzy vision and vertigo, you recover and ungracefully work your way out from under the dead orc. You then start to drag the body to were you want it, but it feels like taking forever. You then dip three bolts in the spider poison and get into position to attack the two sleeping Orcs.
Everyone else, you hear something heavy fall over above you in the tower. Not loud, but you are close and paying attention to anything unusual. This is followed by silence and then something gets dragged across stone.

You would know that the Belladona poison needs to be ingested and the Spider Poison (2d8 on a dc 11 con save) is better suited for injuries.
For the attack:
they are sleeping so your assassin feature applies. If you hit, it will do critical damage.
they are sleeping=incapacitated=advantage on attack
They are prone=disadvantage on ranged attacks
advantage+disadvantage=simple attack role= no sneak attack damage.

Racing for breath - but mostly hoping no one saw her in this position with this creature - she is able to slip out from under it. She drags the flabby, sweaty body overtop the door in the top of the tower. "Gross," she remarks before getting ready. Hope that wasn't too loud, she thinks as she looks at the orc body. She glances at the coiled and ready rope and nods. Nothing for it now, she thinks to herself as she carefully slips a poisoned bolt in the feeder of The Prick.
DM/All - ideally Otrov is looking to shoot the ones under her, not the sleepers - for the very reason you mentioned (disadv.). Plus from where she is and where the description said the sleepers are, I don't think a shot like that is possible. Would the ones below in the courtyard below give the same restrictions on the attack?

I always mess up rolls where Otrov is concerned. Way of reminding myself: Due to Sharpshooter, she ignores disadvantage on ranged attacks. For Assassinate, I am guessing they are not surprised. Any attack on foe that hasn't acted yet is at advantage.
Otrovlines up one of the orcs below and let's the poisoned bolt fly: adv. + sneak - Attack:22Damage:12 IF it hits, orc must roll a CON save (DC11) or take 12 poison damage...

On Margarets command you all jump into action. Bigz reaches the corner of the guardhouse first, from here he casts his spell. One of the Orcs fall over and lands unconscious face first in the grass. A second Orc turns toward his comrade in surprise, the sudden movement nearly causes the bolt that pierces his head to miss, but the impact is so powerful that even the poorly aimed shot kills him instantly. A third one gets hit by a flaming ball of fire, while a single bolt misses it’s target and flies wide (11). Margaret quickly adjusts her aim and takes out the Orc that is injured by the fire with her second shot. Kala struggle a little bit to climb through the breach in the wall and falls a little bit behind.
Two of the Orcs in the north east corner are dead, one is unconscious (5) and one (10) is alarmed that something is going on.
The conscious one seems to be confused and slow to react, you all go before him in initiativ.
Otrov:
A critical hit means you double all the damage dice rolls that are directly linked to the attack. In this case that would be the d8 from the weapon and the 2d6 from sneak attack. So in total that would be 2d8+4d6+dex mod+poison damage. But he is dead dead anyway.
Also you forgot to hide again, so right now you are looking down into the courtyard and the Orc down there can see you.
