If a monster possesses a magic item, you'll want to adjust their stats, saves and/or attacks accordingly, and yeah, you'll need to adjust the creature's stat block manually.
You can create the specific monster NPC as a copy of the "standard" monster, and just change what you need.

If you have already created the statblock for your vampire then just edit that one instead.
Then add the attacks under the Action section, adjust anything else that needs adjusting because of the magic weapon, and then [SAVE CHANGES]. You will likely want to at least familiarize yourself with how to code stuff for the dice roller:
